(color online) Overview of sub-tomogram averaging. Sub-tomograms are extracted from the tomogram. Each sub-tomogram contains a “repeating particle” of interest. Sub-tomograms are rotationally and translationally aligned against a reference. Then, the aligned sub-tomograms are averaged to generate a new reference. Then, the new reference is used for alignment of the sub-tomograms again. This procedure is repeated until the reference stabilizes. Modified with permission from Briggs et al., 2013.
